Fluorescence Test Plate

OverviewThe Fluorescence Test Plate consists of an anodized aluminum body in a microplate format, with Inert Organic Fluorescent Compounds embedded in polymerized matrix. It is designed to be used on BioTek’s FLx800™ and Synergy™ HT series of Multi-Detection Readers. This multi-functional test plate is ideal for GLP compliance while reducing the need for tedious “wet” testing.
General Layout
Three different fluorescent labels are used: green, blue and red. A total of 84 wells with different properties are available on the plate. Most of the tests are based on the green label, using the standard fluorescein filter set (485/20 nm – 528/20 nm). The blue and red labels are provided for reference if additional testing is required eliminating the need to purchase multiple plates.
Tested Parameters
The Fluorescence test plate checks a series of parameters. Top and bottom optics can be checked by turning the plate upside down and repeating the reading sequence.
- Mechanical alignment of the carrier with the optics: this test shows that the result obtained on a sample will not depend on its position on the plate (the relative position of the fiber optics over the well should always be the same).
- Cross talk: this test shows that one or several high positive samples will have no, or very little effect, on the reading of an adjacent negative sample.
- Signal-to-noise ratio: this test shows the ability of the reader to discriminate low fluorescence signal from background.
- Linearity: checks that the relation between the current reading results and the plate’s “Nominal Values” is linear in the operating range of the reader.
- Precision: checks that repeated readings of the same concentration give results with low coefficient of variations. This is checked for both high wells and low wells on the test plate.
Each plate comes with serialized “Nominal Values” obtained on a factory-certified reader, and with an Excel™ spreadsheet that automatically performs data reduction and pass/fail analysis.
